I don't think that you're going to find any online gambling sites that take American Express.
Most, if not all of them, require that money be placed into an online escrow service such as netteller or firepay before they will process a deposit.
Credit card companies want nothing to do with online gambling.
Fortunately, most of these escrow sites are quite reputable and reliable.
I use firepay and have had not one complaint.
